The Grecians head into the game high in confidence after Tuesday night's 6-1 win against Colchester United at St James Park, while Gillingham were 2-1 victors against AFC Wimbledon at the Priestfield.
Speaking ahead of the clash, City boss Matt Taylor says he is looking forward to the challenge of the League One side.
"They're a good side with an experienced manager at this level. This is a cup tie we're relishing and one we're looking forward to, especially with the confidence boost following on from Tuesday's performance.
